Normally a template is associated with just one layout and the correct layout will be selected for you when you select a template. However, you may have more than one layout which would work for a given template so the "Select Layout" button at the top of the Compose screen will reveal a popup allowing you to select a layout from the available list.
Unlike letters and emails which have "regions", there is only one text area on a label, and perhaps one image area as well, though not all label layouts have images on them. If you switch to a layout which is different from the one linked to your label template, be sure to preview your labels to make sure that the image is appearing when you expect it to.
